What is Neutral Red?
Neutral red, dark green powder obtained from 50% ethanol. Soluble in water or alcohol, red in color. Its solubility in water, absolute ethanol, and ethylene glycol-ether is 4%, 1.8%, and 3.75% respectively, and it is insoluble in xylene. LD50142mg/kg (white mouse). Appearance is dark green crystalline powder. It is dark red when dissolved in water and magenta with fluorescence when dissolved in absolute ethanol. The solubility is 4.0wt% in water, 1.8wt% in absolute ethanol, 3.0wt% in ethylene glycol, and 3.75wt% in ethylene glycol ether. It is almost insoluble in xylene, and the absorption wavelength (50% alcohol) is 540nm.
Neutral red is a weakly alkaline pH indicator with a discoloration range of pH 6.4 to 8.0 (from red to yellow). In a neutral or slightly alkaline environment, the living cells of plants can absorb a large amount of neutral red and secrete it into the vacuole. Since the vacuole is generally acidic, the neutral red that enters the vacuole will dissociate into a large amount of cations make it appear cherry red. In this case, the protoplasm and cell wall are generally not colored; dead cells due to the degeneration and coagulation of the protoplasm, the cell fluid cannot be maintained in the vacuoles. Therefore, after staining with neutral red, vacuole coloring will not occur. On the contrary, , the neutral red cation combines with the protoplasm and cell nucleus with a certain negative charge, causing the protoplasm and cell nucleus to be stained.

Add 200 grams of N, N-dimethylaniline to a 2000ml beaker equipped with a stirrer and thermometer. While stirring, slowly add 600 grams of industrial concentrated hydrochloric acid (concentration of 37 to 38wt%). After cooling to room temperature, add Finely crush the ice cubes to reduce the temperature to below 5°C, then slowly add 133 grams of sodium nitrite solution to 250 grams of aqueous solution while stirring. The yield is 56% of the theoretical amount, the purity is ≥97%, the appearance is dark green crystalline powder, and the pH color range is 6.8 (red) to 8.0 (orange) Yellow), sulfated ash content ≤3%, weight loss on drying at 105°C ≤3%.
